Successful treatment of cutaneous leishmaniasis with intralesional aminolevulinic acid photodynamic therapy

Leishmaniasis is a protozoan infectious disease that often affects the skin and may acquire a chronic and difficult to treat course. Topical photodynamic therapy (PDT) is a novel treatment which involves the selective uptake of a photosensitizing agent. Exposure to an appropriate light source in the presence of oxygen leads to formation of reactive oxygen species and destruction of the target cells. We report on the successful treatment of a 69-year-old patient with a relapse of long-standing cutaneous leishmaniasis using intralesional aminolevulinic acid-PDT.

Protection against ultraviolet A-induced oxidative damage in normal human epidermal keratinocytes under post-menopausal conditions by an ultraviolet A-activated caged-iron chelator: a pilot study

Background/purpose: Human skin is constantly exposed to ultraviolet A (UVA), which can generate reactive oxygen species and cause iron release from ferritin, leading to oxidative damage in biomolecules. This is particularly true in post-menopausal skin due to an increase in iron as a result of menopause. As iron is generally released through desquamation, the skin becomes a main portal for the release of excess iron in this age group. In the present study, we examined a strategy for controlling UVA- and iron-induced oxidative stress in skin using a keratinocyte ...

Segmental vitiligo and generalized vitiligo are in general considered to be separate entities. The etiopathogenesis of segmental vitiligo remains unclear, although several hypotheses have been put forward including mainly neuronal mechanisms. The typical association with other auto-immune diseases as seen in generalized vitiligo, seems to be significantly less in segmental vitiligo, although recent insights point towards a possible immune mediated overlap between the two subtypes.

In this article, we describe a case with simultaneous presence of segmental vitiligo, alopecia areata, psoriasis and a halo nevus. To our knowledge, this is the ...

Epigenetics and psoriasis

Abstract

Increasing evidence indicates that epigenetic mechanisms are involved in the pathogenesis of autoimmune rheumatic diseases such as systemic lupus erythematosus (SLE) and rheumatoid arthritis (RA). The pathogenesis of the organ-specific autoimmune disease psoriasis, however, remains poorly understood. Recent studies have shown that aberrant epigenetic patterns are present in patients with psoriasis, suggesting that epigenetic mechanisms may also be involved in this disease. Herein, we review relevant epigenetic findings associated with psoriasis, and provide clues and directions for further studies in this field.

Repeated exposure of human fibroblasts to UVR induces secretion of stem cell factor and senescence

Abstract

Background  Some of chronic hyperpigmentary diseases, such as melasma, induced by multiple factors including chronic sunlight exposure, can recur even after chemical epidermal removal. Dermal factors may be involved in the pathogenesis of melasma. Changes in dermal fibroblasts resulting from chronic sun exposure might cause melanocytes to synthesize melanin in the epidermis.

Objective  This study aimed at determining the effects of repetitive ultraviolet (UV) radiation on cultured fibroblasts and the secretion of melanogenic factors.

Methods  Cultured human fibroblasts were exposed to ultraviolet A (UVA) or ultraviolet B (UVB) for five consecutive days. ...

Inhibition of benzalkonium chloride-induced skin inflammation in mice by an indol-1-ylpropan-2-one inhibitor of cytosolic phospholipase A2α

Abstract

Background:  Irritant contact dermatitis is a frequent and often underrated problem for which the major efficacious therapy is still local glucocorticoids, although they have the known adverse effects due to their wide spectrum of action.

A more focussed therapeutic strategy would be the inhibition of a key enzyme for biosynthesis of the lipid mediators, cytosolic phospholipase A2α (cPLA2α) in irritant contact dermatitis. We are analysing the pharmacological and biological effects of selective cPLA2α inhibitors.

Objective:  To examine the usefulness of the potent and selective cPLA2α inhibitor 3-(5-carboxypentanoyl)-1-[3-(4-octylphenoxy)-2-oxopropyl]indole-5-carboxylic acid (compound 1) for therapy ...

Description of the natural course and clinical manifestations of ichthyosis with confetti caused by a novel KRT10 mutation

Abstract

Ichthyosis with confetti (IWC) was first described as ichthyosis en confettis in 19841 and subsequently as congenital reticular ichthyosiform erythroderma (CRIE)2, also according to the new classification.3 Patients with IWC suffer from erythroderma, prominent scaling, and palmoplantar keratoderma. Because of the development of hundreds to thousands of pale, normal appearing confetti-like spots during childhood, the disease was named IWC.1,4 Our female index patient was described in 1990 as the 4th patient reported worldwide; at that time she did not show any confetti-like spots.5 So far only 3 further IWC patients ...

Comprehensive cytochrome P450 CYP1A2 gene analysis in French Caucasian familial and sporadic porphyria cutanea tarda patients

Summary

Background:  Porphyria cutanea tarda (PCT), the most frequent type of porphyria, results from decreased uroporphyrinogen decarboxylase (UROD) activity. Two forms of PCT are described: a familial form (fPCT) characterized by inherited decrease of UROD activity in all tissues and a sporadic form (sPCT) characterized by decreased UROD activity in the liver. Cytochrome P450 (CYP1A2) plays a major role in triggering experimental uroporphyria in rodents. It has been suggested that the highly inducible -163A/A genotype of the CYP1A2 gene could confer a heightened risk of porphyria cutanea tarda in patients.

Multiple melanoma susceptibility factors function in a UVR response pathway in skin

Abstract

Exposure to ultraviolet radiation (UVR) and the familial melanoma susceptibility gene p16INK4A are among the major risk factors which have been identified to contribute to the development of melanoma, and also significantly contribute to squamous cell carcinoma. Previously, we have shown that UVR induces p16INK4A the expression in melanoma and keratinocyte cell lines and human skin, but the regulatory mechanisms controlling this expression was unknown. Here we show that UVR induced ERK signalling, mediated by BRAF, regulates p16INK4A expression at the transcriptional, and possibly translational level. This study demonstrates the ...

Using optical coherence tomography for the longitudinal non-invasive evaluation of epidermal thickness in a murine model of chronic skin inflammation

Background

Non-invasive methods are desirable for longitudinal studies examining drug efficacy and disease resolution defined as decreases in epidermal thickness in mouse models of psoriasiform skin disease. This would eliminate the need for either sacrificing animals or collecting serial skin biopsies to evaluate changes in disease progression during an individual study. The quantitation of epidermal thickness using optical coherence tomography (OCT) provides an alternative to traditional histology techniques.

Methods
